DALISPIM is an open-top, inverted light sheet microscope that combines high-speed, high-resolution volumetric imaging with broad sample compatibility. Using patent-pending Depth Adaptive Light Interface (DALI) technology, it enables seamless, scalable imaging of diverse tissue types in standard sample carriers like multiwell plates.

DALI technology unlocks new possibilities

In a typical inverted light-sheet setup, the light paths must be oblique to the surface of the sample carrier, which can result in refractive index (RI) mismatches and optical aberrations.

DALISPIM avoids this by pairing an enclosed RI-matching immersion with a deformable interface (see right), creating an RI-matched light path throughout the z-depth range. This allows DALISPIM to achieve optical coupling with standard sample holders – no immersion baths necessary.

Easy to use and ready for automation

Now you can effortlessly acquire high-quality 3D data from samples in standard holders like petri dishes, well plates, slide glasses, and organ-on-a-chip devices – no immersion bath necessary.

DALISPIM is compatible with live or cleared tissues including intact organs, organoids, biopsies, sections, and even entire model organisms. Its flexible configuration accomodates laterally large plates up to 125 x 85 mm, or custom containers up to 250 x 75 x 6 mm.

High-speed, high-resolution microscopy

DALISPIM captures data with near isotropic resolution at imaging speeds 1-2 orders of magnitude faster than conventional confocal microscopy. Custom-designed optics and patented axial sweeping technology (Dean et al. 2015) deliver exceptional image quality for large-scale imaging.

Axial sweeping allows DALISPIM to generate images with uniformly high resolution in all axes without compromising acquisition speed. Illumination optics scan the beam while synchronized to the camera’s rolling shutter detection, providing homogeneous axial point spread functions (PSFz) across the entire field of view.
